Proteasome inhibitors (PIs) are drugs that bind to and inhibit the protein-degrading enzyme complex called the proteasome. PIs are usually used to treat lymphoid malignancies such as mantle cell lymphoma and multiple myeloma, where they work by inhibiting the breakdown of pro-apoptotic proteins such as p53.
